I wanted to elaborate on it today because groups can often get consensus building wrong.  Teams sometimes believe that the goal of consensus building is to get everyone to fall in love with the decision. It’s not. In fact, the goal isn’t even to get everyone to like the decision. The goal of consensus building is to get everyone to live with the decision. That’s a big difference. Let me try to share an example from one of my favorite business books, “The Abilene Paradox” by Jerry B. Harvey. Side note: I had the pleasure of hearing him tell the story firsthand at a conference many years ago. I’m not sure that I could ever tell it the way Jerry does, so I won’t even try. But do check out the synopsis on Wikipedia … it’s really good. The paradox lies in the perils of groupthink, when a team goes along with a decision even though individuals in the group don’t agree with it. An illustration of the paradox could be the office birthday celebration. It’s someone’s birthday so the tradition is to buy a cake. However, truth be told … the cake really isn’t that good. At some point, someone gets the guts to say something about it. Then the company realizes that everyone hates the cake but was going through the motions because they thought everyone else liked it. That’s the paradox. While the birthday cake example is simple, you can see how this can translate when it comes to bigger decisions. The group makes a decision. Amazon Bedrock Agents helps you accelerate generative AI application development by orchestrating multi-step tasks. Agents use the reasoning capability of foundation models (FMs) to break down user-requested tasks into multiple steps. In addition, they use the developer-provided instruction to create an orchestration plan and then carry out the plan by invoking company APIs and accessing knowledge bases using Retrieval Augmented Generation (RAG) to provide an answer to the user’s request. Building intelligent autonomous agents that effectively handle user queries requires careful planning and robust safeguards. Although FMs continue to improve, they can still produce incorrect outputs, and because agents are complex systems, errors can occur at multiple stages. For example, an agent might select the wrong tool or use correct tools with incorrect parameters. Although Amazon Bedrock agents can self-correct through their reasoning and action (ReAct) strategy, repeated tool execution might be acceptable for non-critical tasks but risky for business-critical operations, such as database modifications. In these sensitive scenarios, human-in-the-loop (HITL) interaction is essential for successful AI agent deployments, encompassing multiple critical touchpoints between humans and automated systems. HITL can take many forms, from end-users approving actions and providing feedback, to subject matter experts reviewing responses offline and agents working alongside customer service representatives. The common thread is maintaining human oversight and using human intelligence to improve agent performance. This human involvement helps establish ground truth, validates agent responses before they go live, and enables continuous learning through feedback loops. MIT News - Artificial intelligence The process of discovering molecules that have the properties needed to create new medicines and materials is cumbersome and expensive, consuming vast computational resources and months of human labor to narrow down the enormous space of potential candidates.Large language models (LLMs) like ChatGPT could streamline this process, but enabling an LLM to understand and reason about the atoms and bonds that form a molecule, the same way it does with words that form sentences, has presented a scientific stumbling block.Researchers from MIT and the MIT-IBM Watson AI Lab created a promising approach that augments an LLM with other machine-learning models known as graph-based models, which are specifically designed for generating and predicting molecular structures.Their method employs a base LLM to interpret natural language queries specifying desired molecular properties. It automatically switches between the base LLM and graph-based AI modules to design the molecule, explain the rationale, and generate a step-by-step plan to synthesize it. It interleaves text, graph, and synthesis step generation, combining words, graphs, and reactions into a common vocabulary for the LLM to consume.When compared to existing LLM-based approaches, this multimodal technique generated molecules that better matched user specifications and were more likely to have a valid synthesis plan, improving the success ratio from 5 percent to 35 percent.It also outperformed LLMs that are more than 10 times its size and that design molecules and synthesis routes only with text-based representations, suggesting multimodality is key to the new system’s success.“This could hopefully be an end-to-end solution where, from start to finish, we would automate the entire process of designing and making a molecule. Repurposing Protein Folding Models for Generation with Latent Diffusion

The Berkeley Artificial Intelligence Research Blog PLAID is a multimodal generative model that simultaneously generates protein 1D sequence and 3D structure, by learning the latent space of protein folding models. The awarding of the 2024 Nobel Prize to AlphaFold2 marks an important moment of recognition for the of AI role in biology. What comes next after protein folding? In PLAID, we develop a method that learns to sample from the latent space of protein folding models to generate new proteins. It can accept compositional function and organism prompts, and can be trained on sequence databases, which are 2-4 orders of magnitude larger than structure databases. Unlike many previous protein structure generative models, PLAID addresses the multimodal co-generation problem setting: simultaneously generating both discrete sequence and continuous all-atom structural coordinates. From structure prediction to real-world drug design Though recent works demonstrate promise for the ability of diffusion models to generate proteins, there still exist limitations of previous models that make them impractical for real-world applications, such as: All-atom generation: Many existing generative models only produce the backbone atoms. To produce the all-atom structure and place the sidechain atoms, we need to know the sequence. This creates a multimodal generation problem that requires simultaneous generation of discrete and continuous modalities. Organism specificity: Proteins biologics intended for human use need to be humanized, to avoid being destroyed by the human immune system. Control specification: Drug discovery and putting it into the hands of patients is a complex process. How can we specify these complex constraints?
